 摘要:目的   探讨药物阿魏酸钠对于激素性股骨头坏死早期的干预作用及其对细胞凋亡的影响。方法   利用改良的马血清加甲强龙的方法成功制备兔激素性股骨头坏死早期的动物模型。80只动物随机分为对照组、模型组和治疗组3组。分别选取激素注射后2、4、8、12周4个时间点,每组随机处死6只动物进行股骨头大体观察、HE染色、TUNEL凋亡染色、透射电镜观察,分析每组各时间点相关指标的变化情况以及各组之间的差异。结果 大体观察、HE染色和透射电镜观察发现,早期股骨头坏死动物模型建立成功;治疗组与模型组相比,股骨头坏死表现明显减轻、细胞凋亡数目明显减少,两组差异有统计学意义(P<0.05)。结论 阿魏酸钠可以有效地减轻骨细胞凋亡,延缓或降低股骨头坏死的发生。
Abstract:
 ABSTRACT: Objective   To explore the interventional effects of sodium ferulate on steroid-induced osteonecrosis of rabbit femoral head at the early stage and on apoptosis of osteocytes. Methods   We constructed the rabbit model of early femoral head necrosis with the modified method of horse serum plus methylprednisolone. A total of 80 adult rabbits were randomly divided into 3 groups: the control group, the model group and the treatment group. After intramuscular injection of methylprednisolone, four time points were selected, namely, 2 weeks, 4 weeks, 8 weeks and 12 weeks. Six rabbits in each group were randomly killed; the femoral head underwent gross observation and changes in relevant indexes at each time point were examined by HE staining, TUNEL and transmission electron microscopy. Results   The animal model of early femoral head necrosis was successfully established. Femoral head necrosis significantly was relieved and the number of apoptotic cells was significantly reduced in the treatment group compared with those in the model group (P<0.05). Conclusion   Sodium ferulate can effectively reduce the incidence of osteocyte apoptosis, delay or decrease femoral head necrosis.
